Currently you cannot remove individual files from the media bank - only add new ones or replace old ones. You can remove all files from the media bank by either dragging a folder that has no media (i.e. no QuickTime files or Quartz Compositions) onto the media bank, or by deleting the MediaLibrary file in your ~/Movies/MixEmergency/ folder.
