You can't save effect sequences to specific videos yet - right now it's intended to be more of a live tool. The "Trigger on Load" option is for triggering longer "one-shot" effect animations - for example, you could have a Ripple effect that fades over 3/4 beats that you want to trigger every-so-often manually (i.e. on an emphasised kick, or at the end of a build-up).
